摘 要:目的观察库血保存天数与血浆胆碱酯酶(cholinesterase,ChE)、红细胞乙酰胆碱酯酶(EAChE)活力水平变化的关系。方法库存血标本资料选取健康志愿供血者全血200ml加ACDB50ml,按库血条件保存,按时逐日每份抽取1ml,测定EAChE。当时留取血浆2ml,存入-40℃冰箱,第21天后同时测定血浆ChE。以抽取标本后,即刻测定结果为对照组资料。EAChE测定采用便携式胆碱酯酶测定仪(SBYY)及其配套试剂板;ChE采用Roch公司的胆碱酯酶试剂盒,BeckManCX9全自动分析仪测定。结果库血保存天数为9d以内的血浆ChE值与对照组比较,差异无显著性(P>0.05);而保存10~12d以后的各组均显著低于对照组(P<0.05),保存天数与血浆ChE值呈负相关(r=-0.9888,P<0.01)、直线回归,y=-67.321x+4255,R2=0.9778;但E-AChE水平无显著下降,各组间方差分析差异均无显著性(P>0.05),库血保存时间与EAChE直线回归方程无显著意义(P>0.05)。结论本研究表明库血中血浆内含一定量ChE、红细胞EAChE;EAChE随保存天数延长,21d内无明显下降,为重症AOPP抢救中选用少浆红细胞提供一定的实验依据。Objective To investigatd the relationship between the storage days of banked blood and the plasma cholinesterase, erythrocyte acetylcholinesterase (E-AChE) levels. Methods Two hundred milliliters of whole blood were collected daily from ten healthy volunteers. The blood was added with 50 milliliter ACD before being stored according to the standard of banked blood. One milliliter of whole blood was collected from each blood sample for quick assay of E-AChE. At the same time, two milliliters of plasma were collected from each blood sample, kept in the refrigerator at - 40℃, and the plasma cholinesterase values were determined at 21 days. The blood samples were measured immediately after collected from blood bank and the results served as control group. The assay of E-AChE was accomplished using portable cholinesterase analyzer (SB-YY) and the analyzer was provided with reagent plate. The plasma cholinesterase value was determined with a kit of the Roch Company and the Beck Man CX9 automatic biochemical analyzer. Results There was no significant difference in plasma cholinesterase value between the banked blood kept within nine days and the control group ( P 〉 0.05), but it was significantly lower in the blood stored for more than ten to twelve days groups than that in the control group ( P 〈 0.05). The preservation days was negatively correlated with the plasma cholinesterase value ( r = - 0.9888, P 〈 0.01 ). The linear regression equation was y = - 67.321x + 4255, R2 = 0.9778. However, no significant decrease was noted in E-AChE level, and no significant difference between blood was observed when analyzed using the analysis of variance ( P 〉 0.05). The linear regression equation of the preservation days of banked blood and E-AChE was not statistically significant ( P 〉 0.05). Conclusion Certain amount of plasma ChE and E-AChE exist in banked blood. The activity of E-AChE is not significantly decreased within 21 days.
