摘  要:目的:研究中介素1-47对大鼠急性心肌缺血和离体缺血再灌注损伤的保护作用。方法:利用皮下注射异丙基肾上腺素(ISO)诱导大鼠心肌缺血损伤和离体心脏缺血再灌注损伤大鼠模型,观察中介素1-47对缺血损伤心脏心功能的影响。结果:与正常对照组比较,ISO组大鼠LVESP、MAP、+LVdp/dtm ax和-LVdp/dtm ax分别低41.1%、19.7%、20%和25.2%(均P<0.01),左室舒张末压(LVEDP)增加了517%(P<0.01)。与单纯ISO组比较,低剂量中介素1-47组大鼠的LVESP、MAP、+LVdp/dtm ax和-LVdp/dtm ax分别高8.9%、19%、8.7%和24%,LV-EDP降低52.8%(均P<0.01);高剂量中介素1-47(20 nmol.kg-1.d-1)治疗组,大鼠的±LVdp/dtm ax分别高26.6%和47.4%,LVEDP降低67%(均P<0.01)。心脏缺血损伤后各浓度的中介素1-47再灌注均可显著改善心脏功能。结论:中介素1-47对心肌缺血损伤具有明显的保护作用。Objective To study the effect of intermedin 1 -47 on cardiac function in isoproterenol induced myocardial injury in rats and ischemia/reperfusion (I/R) in isolated rat hearts. Mothod The myocardial Injury model was induced by isoproterenol in rats and ischemia/repeffusion in isolated rat heart. The mean arterial blood pressure (MBP) , heart rate (HR) , left ventricular systolic pressure (LVSP) , diastolic pressure (LVDP) , maximal velocity of increase and decrease of left ventriclar pressure ( ± dp/dtmax ) was monitored on a BL- 410. Results Compared with the normal control group, LVESP and MAP of the ISO group was decreased 41.1% and 19.7%, respectively, ± dp/dtmax decreased 20% and 25.2%, respectively, LVEDP increased 517%. Compared with the ISO group, ±dp/dtmax increased 8.7% ,24%, respectively,in rats given with the treat of low - dose IMD 1 - 47 group ( 5 nmol·kg^-1·d^-1 ), and LVEDP decreased 52.8%.±dp/ dtmax increased 26.6% and 47.4%, respectively, LVEDP decreased 67% in rats given high dose IMD 1 - 47 group ( 20 nmol·kg^-1·d^-1 ). Cardiac I/R induced a marked inhibition of cardiac function and myocardial injury. Reperfusion with IMD 1 - 47 significantly ameliorated the cardiac function inhibition induced by I/R. Conclusion IMD 1 - 47 has cardioprotective effects against myocardial I/R injury.
