摘 要:目的观察Aβ42亚单位肽疫苗接种SD大鼠产生的抗体与阿尔茨海默(Alzheimer)病人脑海马老年斑结合的情况,以筛选Aβ42亚单位片段. 方法先以Bielschowsky改良法确定有老年斑的AD病人的脑,再将Aβ1~15及Aβ36~42亚单位疫苗和Aβ42全肽疫苗接种于SD大鼠,取最后一次接种大鼠的血清与AD病人的脑切片进行免疫组织化学反应(ABC法).结果Aβ1-15亚单位片段疫苗以及Aβ42全肽疫苗诱导产生的抗血清染出的老年斑形态,以经典型(即成熟型)居多,间或有弥散型的老年斑.老年斑略呈圆形,大小不一,遍布海马的各个区域;有些老年斑则呈疏松的丝网状的球体;室管膜下和脑血管膜也见有Aβ沉积.Aβ36-42亚单位片段疫苗诱导产生的抗血清则未能染出老年斑.结论Aβ1-15亚单位疫苗以及Aβ42全肽疫苗诱导产生的抗血清,能与AD病人脑海马切片的老年斑结合;而Aβ36~42亚单位疫苗诱导产生的抗血清,则不能与AD脑片的老年斑结合.Objective The combination of the antibodies which induced by subunit vaccine of Aβ42 with senile plaques in the hippocampus of patients with Alzheimer disease was observed to screen the subunit vaccine of Aβ42 . Methods At first, Senile plaques in the brain of Alzheimer disease were detected by modified Bieslchowsky method. After finishing inoculation on SD rats by the subunit vaccines of Aβ42, the last serum against Aβ42 was stained with senile plaques by using of advin-biotin-complexes immunohistochemistry. Results The antibodies induced by Aβ42 subunit vaccine and complete Aβ42 vaccine can combine with the senile plaques. The main morphological feature of the staining senile plaques was classic type(or mature type), somewhat it was diffusion type .The senile plaques were distributed in the area of the hippocampus and with spherical bodies in various. Some senile plaques showed retiform sphere. Aβ depositions in subventricular zone and cerebral vessels can he detected by the antibodies induced by Aβ1-15 subunit vaccine and complete Aβ42 vaccine. However, the serum induced by Aβ36-42 subunit vaccine can not combined with the senile plaques. Conclusion The serum induced by Aβ1-15 subunit vaccine and complete Aβ42 vaccine can combine with the senile plaques,but it does not happen in the serum induced by Aβ36-42, subunit vaccine.
