摘  要:目的探讨海洛因依赖对大鼠胰岛A细胞胰高血糖素免疫反应(Glu-IR)的影响。方法应用免疫组织化学SABC单染法、形态计量及放射免疫测定法,研究大鼠海洛因依赖期间,A细胞面数密度(NA)及血浆胰高血糖素水平的变化。结果与正常组及盐水组比较,A细胞免疫组织化学显色在海洛因依赖组均增强,阳性细胞增多;A细胞的NA在海洛因依赖第38 d组明显增加,P<0.01;血浆胰高血糖素水平在海洛因依赖第10 d、24 d、38d组均高于正常组和盐水组,P<0.01。结论A细胞通过增加细胞数和增强合成分泌胰高血糖素的功能,参与海洛因依赖期间大鼠糖代谢的调节过程。Objective To explore the effect of heroin dependence on the expression and the function of glucagon in islet A cells of rats. Methods The changes of numerical density on area of A cells and level of plasma glucagon of heroin-dependent rats were investigated by means of the immunohistochemical SABC method,image analysis and radioimmunoassay(RIA) . Results As compared with SCG and NCG, it was observed that the number of GIu-IR positive cells increased and the intensity of immunohistochemical staining enhanced in the heroin-dependent groups. The result of image analysis showed that the numerical density on area of A cell apparently increased in the 38-day heroin-dependent group( P 〈 0.01) . The RIA result showed that the level of plasma glucagon were remarkably higher in all heroin dependence groups than that in SCG and NCG. Conclusion The A cells enhanced the synthesis and secretion of glucagons. The above findings indicated that the A cells of islet were involved in the regulation of carbohydrate metabolic process during heroin dependece of rats.
