摘  要:目的探讨在制备的肝损伤模型中人脐血单个核细胞向肝细胞的分化能力。方法采用腹腔注射CCl4和5-氟尿嘧啶的方法制备肝损伤模型,将分离的脐血单个核细胞以2×107/ml经尾静脉注射入裸鼠体内。4周后处死裸鼠取肝组织。以RT-PCR方法对ALB、AFP mRNA的表达进行检测;并用免疫组织化学SP染色检测人HSA、PCNA、ALB的表达情况。结果RT-PCR显示ALB、AFP mRNA在人肝组织移植组中表达呈阳性,而在损伤未移植组和正常对照组表达为阴性;免疫组织化学染色表明,在裸鼠肝实质区域出现人HSA、PCNA和ALB阳性表达细胞。结论人脐血单个核细胞在裸鼠体内确有向肝细胞分化的趋势。Objective To observe whether the human umbilical, cord blood mononuclear cell could differentiate into cells with hepatocyte phenotype in nude mice with liver injury. Methods Freshly isolated human umbilical cord blood cells were transplanted via tail vein of BALB/c-nu/nu mice. ALB and AFP mRNA expressions were determined in liver at 4 w post- transplantation. HSA, PCNA and ALB protein expressions were examined by immunohistochemical analyses. Results Liver injury model with nude mice had been successfully established. Both of ALB and AFP mRNA were detected with RT-PCR and HSA, PCNA and ALB positive staining was found in transplanted group by immunohistochemistry. They remained negative in control groups. Conclusion Human umbilical cord blood MNCs could differentiate into hepatocyte-like cells in nude mice with liver injury.
