摘 要:目的研究孕鼠被动吸烟对新生大鼠肺组织肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)和细胞因子诱生的中性粒细胞趋化因子(cytokine-induced neutrophil chemoattractant,CINC)表达的影响,探讨产前被动吸烟对新生大鼠肺损伤的机制。方法分别将SD大鼠于妊娠第1、8和15天置于烟箱内,每天被动吸烟2h建立孕鼠被动吸烟动物模型,孕鼠每天置于无烟箱内2h为对照组,于妊娠第20天剖宫取胎鼠测体重,采用逆转录聚合酶链反应(RT-PCR)技术和酶联免疫吸附(ELISA)法检测新生鼠肺组织TNF-α、CINC mRNAs的表达和TNF-α的蛋白水平。结果不同孕期被动吸烟SD大鼠所产新生鼠体重较对照组明显下降(P<0.05或0.01),肺组织TNF-αmRNA的表达与对照组比较明显增强(P<0.01),肺组织TNF-α蛋白含量明显增加(P<0.05或0.01);孕鼠妊娠早、中期被动吸烟所产新生鼠肺组织CINC mRNA表达与对照组和晚期组比较明显增强(P<0.01)。结论孕鼠被动吸烟可影响新生大鼠肺组织炎性介质TNF-α和CINC的表达,这可能是引起新生鼠肺损伤的重要原因之一。Objective To determine the effect of passive smoking during pregnancy on the expression of tumor necrosis factor-α (TNF-α) and cytokine-induced neutrophil chemoattractant (CINC) in the lungs of neonatal rats. Methods Sprague Dawley rats were exposed to sidestream cigarette smoke on the 1, 8 and 15 day after pregnancy for a total of 2 h each day. Those rats who were put into the box without smoke exposure were as control. The neonatal rats were delivered by cesarean section on the 20th day of gestation and birth weight was measured. Reverse transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunoadsorbent assay (ELISA) was performed to evaluate the expression of TNF-α.CINC mRNAs and TNF-α protein. Results The birth weight of neonatal rats born from those mothers who were exposed to sidestream cigarette smoke was significantly lower than that of control gruop(P〈0. 05 or 0.01). The relative amount of TNF-α mRNA expression and TNF-α protein level were also significantly higher than that of control(P〈0.01, P〈0.05 or 0.01). The expression of CINC mRNA in the lung tissue of newborn rats in the passive smoking group at the early and mid-term pregnancy was significantly greater than that of the control group and those exposed to smoke in later stage during pregnancy(P〈0.01). Conclusion Prenatal exposure to sidestream smoke up-regulates the transcription of TNF-α and CINC genes in the lungs of neonatal rats, which may injure the lung tissues.
