摘 要:目的探讨低氧条件下对培养肾小管上皮细胞(HKCs)表达血管内皮生长因子(VEGF)的影响。方法在3%氧浓度下培养肾小管上皮细胞,用细胞生长曲线及四甲基偶氮唑盐(MTT)比色试验研究细胞活力;用原位末端标记(TUNEL)法检测细胞凋亡情况;分别用逆转录聚合酶链反应(RT-PCR)和免疫细胞化学方法检测HKCs在正常和低氧条件下,VEGF mRNA及其蛋白质的表达。结果MTT比色试验较细胞生长曲线更敏感地检测出低氧对细胞代谢的影响,用TUNEL法可检测细胞凋亡率随低氧时间延长而增加,低氧条件下HKCs表达VEGF并无明显增高。结论低氧条件下,检测不到HKCs的VEGF表达增加,可能与低氧诱导HKCs凋亡和代谢率降低有关。Objective To investigate the alternative expression of vascular endothelial growth factor (VEGF) mRNA and protein in human tubular epithelial (HKCs) induced by hypoxia. Methods Human tubular epithelial cell line was cultured in vitro with DMEM/F12 1:1 medium under hypoxia (3% O2) or norm oxygen (control group). HKCs apoptosis was detected by terminal transferase-mediated dUTP nick end-labeling (TUNEL). The expression of VEGF protein was detected by immunohistochemistry and the expression of VEGF mRNA was detected by transcription-polymerase chain reaction (RT-PCR). Results Hypoxia induces HKCs apoptosis rate time-dependently increased. Immunohistochemistry and RT-PCR did not show increasing expression of VEGF mRNA and protein in HKCs under hypoxia condition compared with control group (P〉 0.05). Conclusion Synthesis of VEGF mRNA and protein is not increased in HKCs stimulated by hypoxia. It mayhe due to that hypoxia can induce apoptosis of HKCs.
