摘 要:目的探讨高迁移率族蛋白B1(HMGB1)在烫伤中的表达规律及其对烫伤延迟复苏大鼠脾淋巴细胞周期的影响。方法103只大鼠随机分为正常对照组(n=7)、假伤组(n=32)、烫伤组(n=32)和丙酮酸乙酯(EP)液(3·23mg/ml)治疗组(n=32),后3组分别在伤后第1、3、5、7天活杀,采用Western blot检测血浆HMGB1水平,应用流式细胞仪检测细胞周期。结果大鼠烫伤后1、3、5天血浆HMGB1水平显著升高(P<0·05或P<0·01)。与假伤组比较,烫伤后1、3天G1期淋巴细胞明显增加,同时G2+S期细胞百分比明显减少。EP组,伤后第1天G1期淋巴细胞百分比明显减少(P<0·05),G2+S期细胞比例明显增加(P<0·05)。结论HMGB1对严重烫伤后脾淋巴细胞周期具有明显影响,参与了烧伤延迟复苏后免疫功能紊乱的发病过程。Objective To investigate the change in high mobility group box l protein (HMGB1) levels after scald injury and its effect on splenic lymph cell cycle in rats. Methods 103 male Wistar rats were randomly divided into normal control group (n=7), sham-scald group (n=32), scald group (n=32), and ethyl pyruvate (EP) treatment group (n=32), and they were sacrificed on the 1st, 3rd, 5th and 7th day after a scald injury. The concentration of HMGB1 in plasma was assessed by Western blot, and splenic cells were collected to determine the cell-cycle change with flow cytometry. Results Plasma HMGB1 levels were increased obviously on days 1-5 after scald (P〈0.05 or 0.01). Compared to sham-scald group, an increase in the proportion of sub-G1 splenic cells was concomitant with a marked reduction in the percentage of cells in the sub-G2+S phases on days 1-3 after scald. The inhibition of HMGB1 release by EP was found to result in a decrease in the percentage of cells in the sub-G1 phases and an increase in the percentage of cells in the sub-G2+S phases on days 1 after scald (P〈0.05). Conclusion The results showed that HMGB1 imparted an obvious influence on cell cycle of splenocytes in rats after delayed resuscitation of scald injury, and it might play a role in the pathogenesis of immunologic dysfunction secondary to a major scald injury.
