摘 要:目的观察房室结细胞体外分散培养及形态学特征。方法取新生3 d乳兔房室结组织采用差速贴壁分离技术加机械破坏非心肌细胞法进行原代细胞培养,观察记录各类培养细胞的生长状况,并进行细胞化学染色。结果在培养的房室结细胞中,至少有4种自发性收缩细胞及少量的不规则细胞和多边形细胞。其中梭形细胞最多,圆形细胞和三角形细胞次之,多角形细胞最少;多角形细胞和圆形细胞收缩频率最高,其次为梭形细胞,三角形细胞收缩频率最低。细胞化学染色显示:三角形细胞肌原纤维最丰富,其次为梭形细胞,多角形细胞和圆形细胞肌原纤维最少。结论兔房室结由多种形态不同的细胞构成,多角形细胞和圆形细胞可能是起搏细胞,梭形细胞可能是过渡细胞,三角形细胞可能是普通心肌细胞。Objective To observe primary cultured cells of atrioventricular node(AVN) in detail. Methods The neonatal rabbit AVN cells were cultured and purified with the method of differential attachment and mechanical destructive treatment. The cultured cells were identified by morphology, spontaneously contracting frequency and cytochemical staining. Results Among the cultured cells of AVN, at least four distinctly different types of spontaneously contracting cells were observed , as well as a few irregular cells and polygonal cells. The greatest proportion of the contracting cells were spindle cells and the second group were round and triangle cells and the last group were polyhornal cells. The frequency of spontaneous contraction was the highest in the polyhornal and round cells, inferior to which was the spindle cells, and the frequency of spontaneous contraction was the lowest in the triangle cells. Cytochemical staining showed that the myofibrillae content in the cytoplasm was triangle cells〉 spindle cells〉 polyhornal and round cells. Conclusion The AVN of rabbit was made up by different cells. The polyhornal and round cells may be pacemaker cells; the spindle cells might be transitional cells, and the triangle cells may be myocardial cells.
