摘  要:目的观察大鼠弥漫性轴索损伤(DAI)后不同时间一氧化氮合酶(NOS)在脑皮质、海马及脑干的表达规律,进一步阐明脑损伤后继发性轴索损伤的发生机制。方法参照Marmarou的方法复制大鼠DAI模型,经Gless染色对脑白质、胼胝体、脑干轴索损伤进行形态学观察,应用免疫组织化学方法对不同时间脑皮质、海马、脑干NOS表达规律进行定量观察。结果经Gless染色在伤后0.5 h可见轴索损伤的形态学变化,12 h见明显的轴索收缩球,1~3 d收缩球明显增多,3 d后出现恢复期变化,NOS表达在伤后1 h开始增加,12 h^1 d达到高峰,以后逐渐下降,至10 d仍较基础表达水平为高。结论轴索损伤的形态学变化与伤后不同时间NOS表达的强度有明显的相关关系,表明DAI后NOS参与了继发性DAI的形成过程,为法医学对DAI损伤时间的推断提供新的参考指标。Objective To investigate the changes of nitric oxide synthase (NOS) expression and further elucidate the mechanisms of secondary axonai injury after brain injury. Methods The experimental models of diffuse axonal injury (DAI) were made by the method of Marmarou. The Gless staining was used to observe the morphological changes of axonal damage and the expression of NOS in white matter of the cerebral hemispheres, hippocampus and brainstem was detected by immunohistochemical method. Results Diffuse axonal damage was found at 0.5 h after injury. The retraction ball of axons was observed at 12 h, and increased in the number on the day 1 and 3, and morphological changes of recovery occurred after 3 days. The expression of NOS was increased at first hour after injury, peaked at 12 h to 1 day, and then gradually decreased, but remained above control levels till 10th day. Conclusion The degree of morphological damages of axons is depended on the time-dependent expression of NOS, indicating that NOS might take part in the process of secondary axonal injury.
