摘  要:目的:应用荧光素双标记和不脱钙骨切片,以骨计量学指标对宿主骨变化进行组织学和动态计量分析,以进一步探明不同胚胎起源骨质贴附移植后宿主骨的变化。方法:实验于2002-07/2003-03在军事医学科学院国家二级动物实验室完成。选取新西兰兔10只,分别截取5mm×10mm大小的颅骨(膜内成骨)和髂骨(软骨成骨)各一块,植入兔口鼻部骨膜下,于术后第33,41天分别肌注四环素50mg/(kg·次),进行荧光素双标记,术后第45天处死动物,将移植骨与宿主骨一同取下,制成5μm的不脱钙骨切片,保留平片,其余行Giemsa和VonKossa染色。选用类骨质宽度、骨矿化沉积率指标,对宿主骨变化进行组织学观察和计量学分析。结果:实验纳入新西兰兔10只,全部进入结果分析。①膜状骨和软骨成骨贴附移植后组织学观察:膜状骨移植区宿主骨侧可见明显的成骨细胞排列,宿主骨内及界面类骨质沉积部位较多,可见明显的荧光双标记线及标记区,荧光双标记线距宽。而软骨成骨移植区宿主骨侧可见部分成骨细胞活动,类骨质沉积及荧光标记区少,双标计线窄,增生活跃程度明显较低。②膜状骨和软骨成骨贴附移植后宿主骨区骨计量学的比较:膜状骨移植宿主骨区的骨矿化沉积率、类骨质宽度均明显优于软骨成骨移植区宿主骨区[(3.19±0.37),(2.18±0.36);(14.35±3.48),(5.92±1.68);P均<0.01]。结论:不同胚胎起源骨质移植后的宿主骨,在骨质修复再生速度与成骨量上产生了明显不同的变化,膜内成骨优于软骨成骨,提示膜状骨在口腔颌面部应用时对宿主骨有着更为明显的诱导成骨作用。AIM: To conduct histological and dynamic metric analysis of host bone with bone metric indexes by using the fluorescein double labeled and nondecalcificated bone.section, so as to further explore the changes of host bone after different embryo-derived bone matrix attached transplantation. METHODS: The experiment was carried out in the national grade 2 animal laboratory of Academy of Military Medical Sciences from July 2002 to March 2003. Ten New Zealand rabbits were used, skull (intramembranous ossification) of iliac bone (cartilage bone) of 5 mm×10 mm was removed respectively, and then implanted into oral and nas. subperiosteum, they were treated with intramuscular injection of tetracycline (50 mg/kg per time) on the 33^rd and 4^st days after surgery, and then were fluorescein double labeled, the animals were killed on the 45 days, the grafted bone and host bone were taken together, and prepared into 5μm non-decalcificated bone section, the plain fil was reserved, and others were treated with Giemsa and on Kossa staining. The width of osteoid and the deposition rate of bone mineralization were applied in the histological observation and metric analysis of the changes of host bone. RESULTS: All the 10 New Zealand rabbits were involved in the analysis of results. ① Histological observation after the membranous bone and cartilage bone attached transplantation: In the grafted region of membranous bone, obvious arrangement could be observed on the side of host bone, there were more deposited sites of osteoid inside and on the interface of host bone, obvious fluorescence double labeled line and region could be observed, and the distance of the labeled lines was wide. In the grafted region of cartilage bone: activities of some osteoblasts could be observed on the side of host bone, the deposition of osteoid and fluorescence labeled region were fewer, the double labeled lines were narrow, and the active degree of proliferation was obviously lower. ② Bone metric comparison in host bone region
