摘 要:目的:探讨TLR9在CpG寡脱氧核苷酸(CpG-ODN)种属特异性免疫效应中的意义。方法:CpG-ODN体外刺激人、猕猴、小鼠外周血单个核细胞(PBMC),ELISA测培养液IFN-α含量;RT-PCR检测PBMC中TLR9的表达水平。结果:CpG-ODN2216有效诱导人PBMC分泌IFN-α,而对猕猴和小鼠却无明显的免疫刺激活性;TLR9在人PBMC中均有表达,并且在CpG-ODN介导激活的人PBMC中表达上调;TLR9在猕猴PBMC中不表达。结论:TLR9是构成CpG-ODN种属特异性的重要分子基础。Objective To determine the role of Toll-like receptor 9 (TLR9) in synthetic oligodeoxynucleotides (ODN) containing unmethylated CpG dinucleotides (CpG-ODN) -induced species-specific immune responses. Methods CpG-ODN was co-cultured with peripheral blood mononuclear cells (PBMC) of humans, macaques and mice. The IFN-α in the supernatant was measured by ELISA. The reverse transcription PCR was used to analyze the expression levels of TLR9 mRNA in PBMC. Results CpG-ODN induced high amounts of IFN-α in human PBMC, but had no effect on maeaques and mice. The expression of TLR9 mRNA was observed in all human PBMC, and the levels of TLR9 mRNA were significantly up-regulated with the stimulation of CpG-ODN. We did not observe any expression of TLR9 mRNA in PBMC in macaques. Conclusion TLR9 underlies the moleeular foundation of CpG-ODN-induced species specificity.
