摘  要:目的观察c-ski在创伤和放创复合伤愈合过程中的表达规律,探讨其与创伤愈合和辐射伤组织修复延迟的关系。方法在大鼠创伤和放创复合伤后3,6,9,15d和愈合时,免疫组化和原位杂交检测肉芽组织c-ski表达,5-溴脱氧尿嘧啶核苷(BrdU)掺入检测组织增殖,TdT介导的dUTP缺口末端标记(TUNEL)检测细胞凋亡率。结果创伤后3dc-ski开始表达,9d达到峰值,愈合后显著下降;多种细胞均有高峰度表达,并以成纤维细胞最为显著;辐射侧伤后3~9d的表达显著低于创伤对照侧。辐射导致细胞增殖下降(3~9d)和高峰滞后(15d),与c-ski变化类似。辐射后3~9d的凋亡率显著高于对照侧。结论c-ski可能是一新的组织修复相关基因,它可能通过调节增殖和凋亡的平衡促进创伤愈合。辐射导致c-ski表达下降可能是其延缓愈合的原因之一。Objective To study the expression regularity of c-ski during healing of trauma and radiation-impaired wound and evaluate its roles and relationship with delayed tissue repair. Methods Rat model of wound healing and impaired wound healing was induced bv local soft-X ray. C-ski expression in the granulation tissues was detected by immunohistochemistry- and in situ hybridization at days 3, 6,9 and 15 and at wound healing after trauma and radiation-impaired wounds. Cell proliferation and apoptosis were respectively evaluated by BrdU incorpration and TUNEL methods. Results C-ski began to express at post-wound day 3, reached its peak at day 9 and significantly decreased when wound was healed, c-ski protein was stained in fibroblast, endothelial cell, epithelial cell and hair follicle, especially dominant in fibroblast. Compared to the control side, radiation side showed lower expression of c-ski at post-wound days 3-9. Radiation induced cell proliferation to decrease at days 3-9 and the expression peak to lag at day 15. The apoptosis index was significantly higher on the radiation side at days 3-9 than that of control side. Conclusions C-ski may be a novel tissue-repair-retated gene thai may promote wound healing by balaneing cell proliferation and apoplosis. Radiation can decrease c-ski expre.ssion, which may be responsible for delayed wound healing.
