摘  要:目的:研究白藜芦醇(RSV)对小鼠T淋巴细胞增殖及细胞因子分泌的影响,为阐明其免疫抑制作用提供实验依据。方法:无菌分离小鼠淋巴结细胞,加入不同浓度的RSV预先孵育1h,用多克隆刺激剂佛波醇酯和离子霉素刺激T细胞活化增殖。采用3H-TdR掺入法检测RSV对T细胞增殖的影响;用RT-PCR检测IL-2及IFN-γmRNA的表达;用胞内细胞因子染色法分析IL-2和IFN-γ的分泌。结果:RSV能抑制T细胞增殖,也能在mRNA水平及蛋白水平上抑制IL-2及IFN-γ的分泌。结论:RSV对T细胞的免疫抑制作用,可能与其抑制T细胞的活化、增殖和细胞因子的分泌有关。AIM: To study the effects of resveratrol (RSV) on the activation, proliferation and cytokine expression of murine T lymphocytes. METHODS: Murine T lymphocytes were isolated from lymph nodes and cultured in vitro. The lymphocytes were pre-treated with RSV for I h prior to activation with PDB plus ionomycin. 3H-TdR incorpo- ration was used to detect T cell proliferation. IL-2 and IFN-γ mRNA were detected by RT-PCR, IL-2 and IFN-γ proteins were detected by intracellular cytokine staining with flow cytometry. RESULTS: RSV can inhibit the proliferation and IL- 2 and IFN-γ proteins expression of T lymphocytes. CON- CLUSION: The immunosuppressive action of RSV may be related with its inhibitory effects on T cell activation, proliferation and cytokine expression.
