摘 要:目的:探讨枸杞多糖对兔动脉粥样硬化内皮细胞功能障碍、炎症反应的影响及机制。方法:将36只雄兔随机 分为3组,对照组(12只),喂普通饲料;模型组(12只),普通饲料加1.5%胆固醇喂养;枸杞多糖(Lycium Barbarum Polysaccharide,LBP)组,普通饲料加1.5%胆固醇喂养,同时按体重腹腔注射LBP(3.0mg/kg);10周后,测定各组空腹血清 三脂酰甘油、总胆固醇、高密度脂蛋白胆固醇、一氧化氮、内皮素、丙二醛、C反应蛋白含量以及超氧化物歧化酶活性,计 算主动脉内膜粥样斑块面积并观察主动脉形态学变化。结果:模型组与对照组比较,TG、TCh、NO、MDA、ET-1、CRP明显 升高(P<0.01),HDL-C/TCh、SOD活性明显下降(P<0.01),主动脉内膜粥样斑块面积较大;而LBP组与模型组比较,TG、 CRP、NO、ET-1、MDA明显下降(P<0.01),TCh无明显变化,HDL-C/TCh比值、SOD活性明显升高(P<0.01),主动脉内膜粥 样斑块面积明显减少(P<0.01)。结论:枸杞多糖对动脉粥样硬化有明显的预防和治疗作用,其机制可能与其抗氧化、免疫 调节、提高HDL-C/TCh有关。Objective: To investigate the influence and the mechanism of Lyeium Barbarum polysaeeharide on the function of endothelial cells and inflammatory reaction in atherosclerosis rabbits. Methods: Thirty-six healthy New Zealand white rabbits were randomly divided into three groups,control group was fed with normal diet for 10weeks, model group was fed with 1.5% cholesterol diet, LBP group was fed with 1.5% cholesterol plus 3.0mg/kg LBP. After ten weeks' experiment, the concentration of each group's Triglyeeride, Total Cholesterol, High Density Lipoprotein Cholesterol, C Reactive Protein, Nitric oxide, Maleie Dialdehyde and Endothelin-1 were measured, so was the activity of superoxide dismutase. Results: Compared with the normal control group, model groups's TG、TCh、NO、MDA、CRP and ET-1 increased significantly(P〈0.01), aetivity of SOD and HDL-C/Tch remarkaboy inereased(P〈0.01). While compared with the model group, the LBP group's TG、TCh、NO、MDA, CRP and ET-1 significantly lowered (P〈0.01), while the activity of SOD and HDL-C/Tehremarkably increased (P〈0.01). Conclusion: LBP can effectively accentuating the activity of SOD against oxidation,can regulate immunity, and can evaluate the radio of HDL-C/TCh, to protect endothelial function against atheroselerosis, so LBP is a potential drug in treating and preventing a atheroselerosis.
