摘  要:目的考察从含阿魏酸的小鼠血浆中提取分离微量阿魏酸并制成HPLC微量检测样品的不同方法。方法在小鼠血浆中外加一定量的阿魏酸,分别用甲醇法、乙醇法、加热法提取分离小鼠血浆中微量阿魏酸并进行HPLC检测。结果用乙醇法提取分离小鼠血浆中微量阿魏酸的方法最适用于HPLC检测。结论乙醇法简便快速,重现性好,稳定性强,提取分离效果好,是较理想的小鼠血浆中微量阿魏酸提取分离方法;适用于从小鼠血浆中提取分离微量阿魏酸并用于HPLC法测定小鼠体内阿魏酸血药浓度及药代动力学研究。Objective To observe the different methods when microdose of ferulic acid (FA) was extracted and separated from mouse plasma and the microdetermination sample was made for HPLC. Methods Ferulic acid was added into mouse plasma. Then the microdose of FA was extracted and separated respectively by the methods of carbinal, ethanol and heating, and detected with HPLC. Result The method of ethanol was the best one for HPLC detection. Conclusion The method of ethanol is simple, fast, good for repetition, stable and has a better extractive effect. It is an ideal extractive and separate method for extracting microdose of FA from mouse plasma, detecting the plasmatic FA concentration by HPLC and pharmacokinetic research.
