摘  要:目的采用适当的角膜脱细胞方法获得角膜脱细胞基质,并对其进行特性研究。方法采用胰酶-冰冻法对新鲜兔角膜(全层和分层)进行脱细胞处理,再进行组织学检测及生物学特性力学特性、含水率、膨胀率分析。结果用胰酶-冰冻法获得的兔角膜全层和分层脱细胞基质未见细胞成分,细胞外基质的结构未见明显破坏,脱细胞基质在生物力力学特性、含水率、膨胀率上与正常角膜接近,透光率低于新鲜角膜。结论用胰酶-冰冻法获得的脱细胞基质与自然角膜接近,可作为组织工程角膜支架材料和角膜替代物。Objective To obtain acellular corneal martrix(ACCM) via extraction of cell from cornea and to study its biological characteristics. Methods Fresh rabbit cornea were shaken in 0.25% trypsin for 1 hour and were frozen for 3 days, Cornea were stained with HE and their characteristics were examined, Results There were no celluar components in ACCM and there were no distinct differences in mechanical characteristics, water-content rate and expansive rate between ACCM and cornea. The light trasmittance of ACCM was lower compared to cornea. Conclusion ACCM can be used as a biological scaffold for tissue engineering and corneal substitute.
