摘 要:目的研究低氧(2%氧)对成年Wistar大鼠心脏成纤维细胞胶原蛋白合成、总蛋白合成及Ⅰ、Ⅲ型胶原前α肽链mRNA表达的影响。方法分离培养成年Wistar大鼠心脏成纤维细胞,采用液体闪烁计数方法检测心脏成纤维细胞胶原酶敏感的3H-脯氨酸掺入率来观察细胞胶原蛋白合成变化,并检测3H-亮氨酸掺入率观察细胞总蛋白合成的变化,采用RT-PCR检测Ⅰ、Ⅲ型胶原前α肽链mRNA的表达。结果成年Wistar大鼠心脏成纤维细胞在低氧第24、48小时胶原酶敏感的3H-脯氨酸掺入率较常氧组显著增加,分别增加132%(P<0.05)和163%(P<0.01);低氧12h起Ⅰ、Ⅲ型胶原前α肽链mRNA表达显著高于常氧培养的细胞。结论低氧能够直接促进体外培养的成年Wistar大鼠心脏成纤维细胞胶原蛋白合成和Ⅰ、Ⅲ型胶原前α肽链mRNA的表达,提示低氧对心脏成纤维细胞胶原合成代谢的直接调节可能是低氧性心肌纤维化的重要机制。Objective To observe the effects of hypoxia on collagen protein and total protein synthesis and the expression of collagen type Ⅰ and Ⅱ mRNA in cultured cardiac fibroblasts of male adult rat. Methods Cardiac fibroblasts (CFs) were isolated from adult Wistar rat ventricle and cultured in vitro either in normoxic or hypoxic condition. The changes of collagen protein and total protein synthesis were determined by measuring the collagenase sensitive 3H-proline incorporation and the ^3H-leucine incorporation respectively. The changes of expression of pro-α1 (Ⅰ) collagen and pro-α1 (Ⅲ) mRNA were measured by RT-PCR. Results The collage- nase sensitive 3H-proline incorporation of CFs was increased by 132% (P 〈0.05 )and 163% (P 〈0.01 ) after 24-hour and 48-hour exposure to hypoxia (2% O2) respectively. The level of pro-α1 (Ⅰ ) collagen mRNA and pro-α1 (Ⅲ) mRNA was significantly elevated in the cells exposed to hypoxia for 12 h, 24 h, 48 h. Conclusion Hypoxia alone can up-regulate collagen synthesis in cardiac fihroblasts of adult rat, which may he one of the important mechanisms that hypoxic myocardial fibrosis occurs.
