豚鼠海马锥体细胞在视觉分辨任务中放电时相变化的初步研究

摘  要:目的设计有效、简便的锥体细胞放电时相检测程序,并检测豚鼠海马CA1区锥体细胞在视觉分辨任务中放电时相的变化.方法对执行视觉分辨任务的5只豚鼠进行在体单细胞外记录.利用小波变换对豚鼠海马锥体细胞放电信号进行处理,提取单个锥体细胞放电信号,并根据时间关联获得锥体细胞的放电时相.结果海马CA1区锥体细胞在视觉分辨任务前1~5 s的放电时相(172°±1.8°)明显早于任务后6~10 s的放电时相(189°±3.7°)(P<0.01).而任务后1~5 s和11~15 s的放电时相(177°±6.0°,172°±3.5°)则没有明显变化(P>0.05).结论程序可以有效、简便地检测锥体细胞的放电时相.海马CA1区锥体细胞在视觉分辨任务执行过程中的放电时相存在有规律的移动.Objective To detect the change of discharge phase of guinea pig hippocampal CA1 pyramidal cells during visual discriminative task with an effective and convenient program we designed. Methods Five guinea pigs were performed by extracellular single unit recording in vivo when they were performing visual discriminative task. Discharge signals of individual pyramidal cells were extracted from different frequency signals by wavelet transform (WT), which made it feasible to calculate discharge phase of pyramidal cells in terms of time correlation between discharge and θ rhythm. Results The discharge phase of CA1 pyramidal cells in the 1 - to 5 -s interval before visual discriminative task ( 172°± 1.8°) was obviously earlier than that in the 6- to 10-s interval after visual discriminative task ( 189°±3.7° ) ( P 〈 0.01 ), whereas the discharge phases in the 1 - to 5-s and 11 - to 15-s interval after visual discriminative task were of no significant difference (P 〉 0.01 ). Conclusion The program we designed is capable of detecting discharge phase of pyramidal cells. Regular shift of discharge phase of hippocampal CA1 pyramidal cells emerges before and after performing visual discriminative task.
