摘  要:目的评价载药自固化磷酸钙人工骨的抗菌活性和骨诱导活性。方法将自固化磷酸钙人工骨(CPC)与重组人骨形成蛋白(rhBMP)-2和亚胺培南/西司他丁钠(商品名:泰能)复合,制成载药自固化磷酸钙人工骨(diCPC);将diCPC置于接种有金黄色葡萄球菌(简称:金葡菌)的培养基中,检测其体外缓释特性及抗菌活性;将diCPC植入小鼠股部肌肉内,以单纯CPC植入作对照,术后2周和4周,通过组织学检查评价其骨诱导活性。结果diCPC在体外长时间持续释放抗生素并抑制金葡菌生长。diCPC植入小鼠股部肌肉2周,局部有大量的软骨细胞分化及软骨基质形成;术后4周,局部有大量新生的编织骨形成。CPC植入后2周和4周,材料周围仅有纤维结缔组织包裹,未见软骨或骨形成迹象。结论diCPC具有良好的抗菌活性和骨诱导活性,是伴有污染或感染骨缺损较理想的修复材料。Objective To explore osteoinductivity and antibiosis of drug-impregnated calcium phosphate cement (diCPC). Methods diCPC was made by putting recombinant human bone morphogenetic protein( rhBMP)-2 and Tienam into calcium phosphate cement (CPC). The sustaining period of effective drug concentration in vitro was detected by inhabiting bacteria, diCPC was implanted into the muscle pouches of mice, and CPC alone was implanted as control. All the animals were killed at intervals of 2,4 weeks after the operation, and the specimens were evaluated by light microscope. Results diCPC sustained a high concentration level of antibiotics in vitro for at least thirty-four days. Cartilage growth around diCPC was observed for two weeks, and woven bone was observed four weeks after the operation. No cartilage or bone was observed around implanted CPC two or four weeks after the operation. Conclusion diCPC possesses a superior osteoindutivity and antibiosis, and it may be ideal as a bone graft substitute for bone defect of contamination or infection.
