摘 要:目的观察人参银杏合剂对大鼠缺氧时的抗氧化神经保护作用。方法参杏合剂由西洋参和银杏叶煎制而成,生药含量0.2g/mL。分别观察低氧、常氧状态下药物合剂对SD大鼠脑组织中谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH-PX)和过氧化氢酶(CAT)活性的影响,并检测药物合剂在体外的抗羟自由基效力。以低压舱减压模拟高原缺氧造成大鼠缺氧脑损害为模型。低氧实验时,SD大鼠分成常氧组、低氧组、药物组。药物于缺氧前48h、24h、1h给药,对照动物给等量的生理盐水(1mL/200g·wt)。结果①缺氧实验结束后,与常氧组比较,低氧暴露组动物脑组织中GSH含量下降,GSH-PX和CAT活性降低,使用参杏合剂后,GSH、GSH-PX、CAT均有不同程度的改善(P<0.05或P<0.01)。②常氧状态下,与生理盐水组比较,药物对上述3指标都有一定的升高作用(P<0.05或P<0.01)。③参杏合剂能直接清除体外实验体系中的羟自由基。结论在本实验条件下,参杏合剂对大鼠缺氧脑组织有一定的抗氧化神经保护作用,药物的这种作用与其提高机体自身的抗氧化能力及其直接的清除自由基作用有关。Objective To observe the anti - oxidative effects of Ginseng - Ginkgo biloba mixture against hypoxic brain in rats. Methods The hypoxic condition was established with a hypobaric chamber. Adult Sprague -Dawley rats were divided into control group, hypoxia group and drug group. The rats were pretreated intraperitoneally at 48 h, 24 h, and 1 h before hypoxia in drug group. The saline was used in other two groups. The hydroxyl radical scavenging activity of drug mixture was measured in vitro using alizarin violet spectrophotometry. Results Compared with normal control group, content of GSH, activities of GSH- PX and CAT decreased significantly in hypoxic group, but they were elevated in the drug group. In normal experiment, compared with control group, content of GSH, activities of GSH- PX and CAT were elevated with drug mixture respectively. In vitro, drug mixture exhibited hydroxyl radical scavenging property. Conclusion The results suggest that Ginseng - Ginkgo biloba mixture plays an anti- oxidative neuroprotective role on rat brain exposed to hypoxia.
