摘 要:目的:研究灌注肝动脉和冲洗胆管方式在冷缺血期对大鼠供肝胆管细胞的影响。方法:各组均以威斯康星大学液(UW液)灌注大鼠供肝门静脉,以不同方式灌注肝动脉和冲洗胆管,在冷保存的不同时间段观察胆管细胞的变化。结果:冷保存4h后,各组光镜下胆管细胞无明显变化。冷保存8h后,UW液灌注大鼠供肝门静脉、肝动脉,经UW液、Ringer's液冲洗胆管的两组胆管细胞变化较其它组变化小,并且两组之间无明显差异。冷保存12h后经UW液灌注大鼠供肝门静脉、肝动脉及冲洗胆管组胆管细胞变化较其它组变化小。结论:灌注肝动脉和冲洗胆管有助胆管细胞保存,8h内只要能够完全清除胆盐或降低胆盐浓度,即可产生相同的保护胆管效果。随着冷保存时间的延长,应用UW液冲洗胆管可以提高供肝胆管细胞的保存效果。Objective To explore the effect of preservation through hepatic artery and washing biliary tract on biliary tract cell of rats donor liver in cold preservation. Methods With portal vein of donor livers perfused through by UW solution in every group, perfuse through hepatic artery and wash biliary tract by different methods, and then observe the change of biliary tract cell on different time in the cold preservation. Results There were no obvious changes under different light microscope after 4 hours in cold preservation. After 8 hours in cold preservation, with portal vein and hepatic artery of donor liver of rats perfused through by UW solution, two groups of biliary tract cell which perfused by UW solution and Ringer's solution changed mostly less than other groups, and there was no obvious difference between them. After 12 hours in cold preservation, the change of the group whose portal vein and hepatic artery perfused through by UW solution and biliary tract washed by UW solution was less. Conclusions Perfusing hepatic artery and washing biliary tract benefit the preservation of biliary tract cell. In 8 hours if you can clear choline or lower the density of choline, you can get the same effect. With the cold preservation lasting, we can get better effect of preservation of biliary tract cell when washing biliary tract by UW solution.
