摘 要:目的:动脉的中膜平滑肌细胞(SMC)向内膜移行和增殖是经皮腔内冠状动脉成形术(PTCA)后再狭窄的主要原因。为探讨水蛭素预防再狭窄的可能性,本文观察了水蛭素对培养的兔胸主动脉SMC增殖的影响。方法:用4只兔的胸主动脉SMC传代培养(5~6代),将水蛭素(1~25μg/ml)加入SMC培养液中,于1、2、3天后进行细胞计数并于6小时后测定氚-胸腺嘧啶脱氧核苷(3H-TdR)参入的放射性强度每分钟次数(cpm)值。求出各组均数及标准差,采用方差分析及q检验进行统计分析。结果:各浓度组的水蛭素均抑制SMC的增殖及对3H-TdR的摄取,且呈剂量依赖性,与对照组相比P<0.05或P<0.01。结论:水蛭素抑制培养的兔动脉SMC的增殖,这为水蛭素可能用于预防以SMC增殖为基础的再狭窄提供实验依据。Objective:Migration of smooth muscle cells(SMCs) from arterial media to intima and proliferation of the intimal SMCs are the main cause of restenosis following successful percutaneous transluminal coronary angioplasty (PTCA).For evaluating the possibility of hirudin on the prevention of restenosis,we examined the effects of hirudin on the proliferation of rabbit aortic SMCs cultivated in vitro. Metheds:5~6th passage culture of aortic SMCs from 4 rabbits was used,hirudin was added into the medium of cultured SMCs in concentrations ranging from 1 to 25μg/ml.The numbers of SMCs were assessed by cell counting at 1,2,3 days and the cpm values of 3H-TdR up -take were determined at 6 hours after hirudin addition. Results:Hirudin (1-25μy/ml)inhibited the proliferation and 3H-TdR up take of SMCs in a dose-dependent manner(p<0.05 or 0.01 vs. control). Conclusion:Hirudin inhibits the proliferation of cultured rabbit aortic SMCs,suggesting the possibility that hirudin can be used to prevent restenosis following PTCA.
