补肾活血与益气活血方药对脑缺血老龄大鼠白介素-8和肿瘤坏死因子的影响

摘　　要：目的探讨缺血性脑损伤病理,比较补肾活血与益气活血方药防治机制,为其临床应用提供依据。方法采用二血管(CCA)阻断方法复制老龄大鼠急性脑缺血模型;观察具有补肾益元、活血通络功效的中药复方益元活血丹及具有益气活血通络功效的中药复方益气活血方对脑缺血老龄大鼠白介素(IL)-8和肿瘤坏死因子(TNF)水平的影响。结果与老龄假手术组比较,老龄模型组血清IL-8、血清及脑组织TNF水平明显升高;与老龄模型组对照,两方药组血清IL-8、血清和脑组织TNF水平降低。结论老龄大鼠急性脑缺血损伤可能在于IL-8、TNF水平升高,加重脑组织缺血损伤;补肾活血和益气活血方药对急性脑缺血防护作用显著,其机制可能在于抑制IL-8、TNF介导的脑组织损害作用;两方药均降低脑缺血损伤血清IL-8水平而起保护作用,但作用机制有所侧重:益气活血方药降低血清IL-8水平较明显。Objective:To study the pathogenic mechanism of ischemic brain damage in aged rats and prevention and cure mechanism of drugs for Tonifying Kidney and Promoting Blood Circulation(YD),for Increasing Qi and Promoting Blood Circulation(YF), to provide experiment's basis in the clinical application for drugs.Methods:Observed effect of drugs for YD and YF in aged rats with cerebral ischemia on IL-8 and TNF.Results:The serium IL-8,while the TNF of serium and brain tissue in the aged model group were elevated that those in the aged control one.Contrasted to the aged model group,the IL-8 and TNF of the two herbs druged groups were decreased.Conclusion:Ischemic brain damage in aged rats may concern with the rising of the IL-8 and TNF.YD and YF could significantly reduce the cerebral damage caused by ischemia.Hence, the functional mechanism might concern with inhibiting the damage if IL-8 and TNF.The protection of the TCM herbs were both rested in depressing the IL-8 and TNF. the YF declined the serium IL-8 level significantly.
