摘 要:目的研究兔骨髓间质成骨细胞(RMSO)与兔肾脏血管内皮细胞(RRVEC)共培养时,1,25-(OH)2D3对细胞活性及碱性磷酸酶(ALP)活性、骨钙素(OCN)含量的影响。方法以相同总细胞数接种24孔细胞培养板,1,25-(OH)2D3作用5d后,MTT比色法测定细胞活性。以相同RM-SO细胞数接种96孔细胞培养板,分别于1,25-(OH)2D3作用3、5、12d时,测定细胞ALP活性、OCN含量。结果各检测时间点1,25-(OH)2D3作用组及与RRVEC共培养组细胞活性、ALP活性和OCN含量均较RMSO单独培养增强(P<0.05)。1,25-(OH)2D3作用于RMSO、RRVEC共培养时,在各检测时间点,细胞活性、ALP活性、OCN含量均进一步增强(P<0.05)。结论1,25-(OH)2D3单独应用可促使RMSO成熟,1,25-(OH)2D3作用于RMSO、RRVEC共培养时,细胞活性、RMSO细胞分化及成骨能力均进一步增强。Objective To evaluate the effect of 1,25-dihydroxyvitamin D3 [1,25-(OH)2D3 ]on coculture of rabbit marrow mesenchymal stem cells(MSC) and renal endothelial cells(EC). Methods With same number of total cell in 24-well tissue culture plate, cell activity was evaluated on the 5th day after use of 1,25-(OH)2D3 and with same number of MS(2 in 96-well tissue culture plate, alkaline phosphatase(ALP) activity and osteocalein(OCN) production were detected on the 3rd, 5th, and 12th day after 1,25-(OH)2I~ application. Results Application of 1,25-(OH)2D3 had the same effect on MSC as co-culture of EC, which all made the cell activity, ALP, OCN increased. When 1,25-(OH) 2D3 applied to co-culture of MSC and EC, all the measurements increased farther. Conclusion 1,25-(OH)2D3 can accelerate the differentiation of MSC and when it is applied to co-culture of MSC and EC, the bone forming ability of MSC is increased further.
