摘  要:目的探讨白细胞介素18(IL18)与异基因小鼠骨髓移植后急性移植物抗宿主病(aGVHD)的关系及作用机制。方法C57BL/6小鼠随机分为A、B、C3组,经60Co照射后,A、B组经尾静脉移植入Balb/c小鼠骨髓细胞,C组移植入C57BL/6小鼠骨髓细胞。移植后,A组注射IL18,B、C组注射NS。测定B、C2组小鼠血清IL18浓度;比较A、B2组小鼠aGVHD的程度,酶联免疫法(ELISA)测定血清中干扰素γ(IFNγ)及白细胞介素10(IL10)的浓度。结果移植后B组小鼠血清IL18的浓度明显高于C组;A组小鼠较B组小鼠的aGVHD严重,血清IFNγ的浓度高,血清IL10的浓度低。结论①aGVHD发生时,血清IL18浓度升高。②IL18可以加重异基因骨髓移植后aGVHD反应。③IL18促进CD4+T细胞向CD4+Th1细胞分化可能是IL18加重aGVHD反应的机制之一。Objective To investigate the role IL-18 in acute graft-versus-host disease (aGVHD) after allogeneie bone marrow transplantation (Allo-BMT) in mice. Methods C57BL/6 recipient mice irradiated with ^60Co were randomly divided into A, B, and C groups. Mice in group A and B were transfused with Balb/c donor cells while mice in group C with C57BL/6 donor cells. After transplantation, recipient mice in group A were treated subcutaneously with 1μg IL-18 daily from day 0 to day 5 while recipient mice in group B and C with NS, separately. In group A and B, the severity of aGVHD was evaluated. Concentrations of IL-18, IFN-γ and IL-10 in serum of mice were measured by enzyme-linked immunoserbent assay (EHSA). Results The concentration of IL-18 in serum of B group was higher than that of C group after transplantation, aGVHD response was more severe in group A than that in group B. The serum concentration of IFN-γ in group A was higher than that in group B, but the serum concentration of IL-10 was lower than that in group B. Conclusion The serum concentration of IL-18 is increased when aGVHD occurred in recipient mice after allogeneic transplantation. IL-18 may aggravate aGVHD response. IL-18 can promote the differentiation of CD4^+ T cells into CD4^+ Th1 cells, which may be one of the mechanisms of aggravating aGVHD.
