摘  要:目的观察兔正畸牙移动过程中牙周组织血小板衍生生长因子AA(platelet-derivedgrowthfactorAA,PDGF-AA)的表达与分布,探讨血小板衍生生长因子-AA在正畸牙移动过程中的作用机制。方法选择35只体重为2.0 ̄2.5kg的健康新西兰大耳白兔,随机分成7组,每组5只,分别为1、3、5、7、14和21d正畸加力组和正常对照组。2%戊巴比妥钠麻醉,兔下切牙常规粘网底颊面管,以镍钛推簧施力40g。采用免疫组织化学染色进行血小板衍生生长因子-AA半定量分析。结果血小板衍生生长因子-AA在正畸加力组牙周组织的表达明显强于正常对照组(P<0.01),7d组为表达高峰期,14、21d组表达减弱。并且同一实验组,血小板衍生生长因子-AA在张力侧表达均高于压力侧(P<0.01)。结论血小板衍生生长因子-AA在兔正畸牙移动过程中的表达明显増强,从而推测血小板衍生生长因子-AA可能参与了正畸骨改建过程的骨形成调节,并发挥着重要作用。[Objective] To investigate the expression and distribution of platelet-derived growth factor AA (PDGF-AA) in the periodontal tissues and analyze the role of PDGF-AA in orthodontic tooth movement. [Methods] 35 white rabbits were randomly divided into 7 groups with 5 rabbits in each group, including experimental group of 1, 3, 5, 7, 14 and 21 days and one control group. After anesthesia with 2% pentobarbitone sodium, orthodontic appliances consisting of a coil spring were fixed between the lower incisors, the force experted at the time of insertion was approximately 40 g. The expression of PDGF-AA was investigated half-quantitatively through immunohistochemical analysis. [Results] The expression of PDGF-AA in the experimental group enhanced apparently compared with that in the control group (P〈0.01). On 7 day group it reached the peak level. On 14 and 21 days group the expression of PDGF-AA declined. And the level of PDGF-AA of the experimental group at tension side was higher than that at pressure side at the same time (P〈0.01). [Conclusion] PDGF-AA participate in the periodontal tissue remodeling during orthodontic tooth movement and play a important role in orthodontic bone formation.
