摘 要:目的建立大黄配方颗粒中大黄酸、大黄素、大黄酚的含量测定方法.方法采用反相高效液相色谱法测定,用Kromasil ODS柱(250 mm×4.6 mm,5 μm),以甲醇-0.1%磷酸溶液(85∶15)为流动相,检测波长254 nm.结果大黄酸、大黄素、大黄酚的线性范围分别为0.010 9~0.218 0 μg、0.011 2~0.224 0 μg和0.011 9~0.238 0 μg;大黄酸平均回收率为100.6%,RSD=0.92%(n=9);大黄素平均回收率为100.5%,RSD=0.85%(n=9);大黄酚平均回收率为100.6%,RSD=1.15%(n=9).结论所用方法测定样品分离效果佳,灵敏度高、重复性好,能有效地控制大黄配方颗粒的质量.Objective To establish a quantitative method for rhein , emodin and chrysophanol in Dahuang Dispensing Granule by RP-HPLC. Methods The sample were separated by Kromasil ODS column( 250 mm× 4.6 mm, 5 μm)with methanol- phosphoric acid 0. 1 %(85 : 15)as mobile phase, detection wavelength at UV 254 nm. Results The linear relationship was the range of rhein within 0. 010 9-0. 218 0μg ,emodin in 0. 011 2-0. 224 0 μg and chrysophanol in 0.011 9-0.238 0μg ,respectively. The average recovery of rhein was 100.6% with RSD of 0.92% (n=9), emodin was 100.5% with RSD of 0.85%(n= 9) and chrysophanol was 100.6% with RSD of 1.15% (n=9) ,respectively. Conclusion The method is available with a good separable efficiency, reproducibility and sensitivity, which can be well controlled the quality of Dahuang Dispensing Granule.
