摘  要:目的研究低氧(2%氧)对成年Wistar大鼠心脏成纤维细胞表型的影响。方法分离培养成年Wistar大鼠心脏成纤维细胞,采用免疫细胞化学染色检测肌成纤维细胞的典型标记———α平滑肌肌动蛋白,用液体闪烁计数方法检测心脏成纤维细胞胶原酶敏感的3H脯氨酸掺入率来观察细胞胶原蛋白合成变化,并检测3H亮氨酸掺入率观察细胞总蛋白合成的变化。结果低氧24h、48h时,成年Wistar大鼠心脏成纤维细胞的α平滑肌肌动蛋白表达较常氧组显著增强;同时细胞胶原蛋白合成速率亦显著增加,在低氧第24h、48h胶原酶敏感的3H脯氨酸掺入率较常氧组分别增加132%(P<0.05)和163%(P<0.01)。结论低氧能够促进成年Wistar大鼠心脏成纤维细胞向肌成纤维细胞表型转换。Objective To observe the effects of hypoxia on phenotype of cultured adult rat cardiac fibroblasts. Methods Cardiac fibroblasts(CFs) were isolated from adult Wistar rat ventricle and cultured in vitro. Then CFs specimens were divided into two groups:control group and hypoxic group, and cultured in normal oxygen condition and low oxygen(2%) tank respectively, α-smooth muscle actin (α-SMA) protein level was determined by immunostaining, collagen protein and total protein synthesis were determined by measuring the collagenase sensitive 3 H-proline incorporation and the 3 H-leucine incorporation after cultured for 24 and 48 h respectively. Results As compared with con- trol group the collagenase sensitive 3 H-proline incorporation of CFs was increased by 132 % (P〈0.05) and 163%(P〈0.01) after 24 h, 48 h hypoxia exposure respectively. Conclusions The results suggest that hypoxia can induce differentiation of adult rat cardiac fibroblasts into myofibroblasts.
