摘 要:热卡限制能使众多实验动物的寿命延长,而且它是唯一延缓哺乳动物衰老的途径。我们用原生动物阴道毛滴虫建立了热卡限制模型。在此研究体系中,该原生动物细胞周期在热卡限制的培养条件下较糖充足的培养条件下延长80%。本研究运用抑制性消减杂交的方法,以期筛选在两种培养条件下差异表达的基因,并从2个cDNA消减杂交文库中分离到15个克隆。经序列分析表明,有9个未知基因,6个已知基因。That calorie restriction (CR) extends lifespan has been observed in most laboratory species, and the only way to slow down aging in mammals. We established a model of CR in the protist Trichomonas vaginalis. In this system, the cell cycle of organism could be extended by reducing the concentration of glucose. Differences in gene expression are likely to explain the cell cycle extension in CRT. vaginalis. To screen differentially expressed genes in the normal and CRT.vaginalis, suppression subtractive hybridization (SSH) was utilized and 15 differentially expressed clones were isolated.Sequence analysis revealed that 9 were unknown genes which did not match to any previously reported genes, the other 6were homologues of known genes.
