摘  要:目的:探讨胰岛素抵抗在非乙醇性脂肪肝(NAFLD)中的作用。方法:8只SD大鼠随机分成正常对照组(6只)和高脂饮食组(42只),正常对照组饲养8周,高脂饮食组分为7组,每隔2周处死1组,测定大鼠的体重,肝脏的湿重,空腹真胰岛素(FIns)、血糖(FBG)、血脂水平,观察肝组织学改变。结果:模型组大鼠喂养8周时出现NAFLD。与正常对照组相比,模型组大鼠的FBG,FIns,TG,高密度脂蛋白胆固醇(HDL-C),胰岛素敏感指数(ISI),肝细胞脂肪变程度与对照组相比均有差异。TG,FIns与肝细胞脂肪变程度成正相关,ISI,HDL-C与肝细胞脂肪变程度成负相关。结论:模型组大鼠存在胰岛素抵抗;胰岛素抵抗在非乙醇性脂肪肝的发生中起重要作用。Objective: To explore the effect of insulin resistance on the pathogenesis of non-alcoholic fatty liver disease(NAFLD). Methods: Forty eight male SD rats were randomly allocated into control group (n = 6 ) fed with standard diet for 8 weeks and model group( n=42 ) induced by high fat diet. The model group was divided into 7 groups ,these rats were sacrificed at the 2,4,6,8,10,12,14wk of the experiment respectively. Body weight, liver weight, fasting blood glucose (FBG) , serum triglyceride (TG) , fasting serum insulin(Fins) level and liver histology were detected. Results:The livers presented the pathology of hepatocyte steatosis at 8 week in rats of model group. In comparison with the control group, there was difference in FBG, Fins, TG, insulin sensitivity index (ISI), HDL-C and the degree of fatty liver in model group. TG and Fins were positive correlation with the degree of hepatocyte steatosis. Conclusion :Insulin resistance generally presents in rats with NAFLD. Insulin resistance may play an important role in the pathogenesis of NAFLD.
