摘 要:目的观察射心通胶囊对急性心肌梗死(AMI)大鼠一氧化氮(NO)、一氧化氮合酶(NOS)的影响,探讨射心通胶囊扩张冠脉、增加缺血心肌供血的机制。方法Wistar大鼠随机分为5组:对照组、心肌梗死模型组、射心通胶囊(低、中、高剂量)3个组,各组每日用药1次,连续5d。用手术结扎法复制大鼠心肌梗死模型,测定血浆中NO浓度;用免疫组化法测定大鼠心肌内皮型一氧化氮合酶(eNOS)活性。结果射心通胶囊3个剂量组均可显著升高心肌梗死大鼠血浆和心肌组织的NO含量(P<0.05),明显提高心肌细胞内eNOS的活性(P<0.05)。结论射心通胶囊可以提高eNOS活性,从而升高NO水平,达到改善心肌缺血的目的。Objective To observe the influence of Shexintong capsule on nitric oxide(NO) and endogenic nitric oxide synthase (eNOS) in model rat of acute myocardial infarction (AMI), furthermore to investigate the mechanism of Shexintong capsule dilated coronary artery and improved the blood offering of myocardium. Methods Wistar rats were randomly divided into five groups: control group, AMI group, Shexintong capsule (low, middle and high dose) groups. Rats were treated with drugs per day and totally five days. Surgery was performed in rats to duplicate the AMI model. We measured the NO concentration in blood plasma and the eNOS activity in myocardium with immunohistochemistry method. Results Three groups of Shexintong capsule could significantly enhance the NO content in plasma and myocardium of AMI rats (P〈0.05), also significantly enhance the NO activities in myocardiocytes (P〈0.05). Conclusion Shexintong capsule could improve the level of NO through enhancing the activity of eNOS, therefore it induced the effect of reducing myocardial ischemia.
