摘 要:应用粘附式细胞激光定量分析及筛选仪(ACAS-570,美国)建立定量研究活细胞内DNA含量和钙离子浓度的实验模型。初步观祭微量直流电刺激对成骨细胞分化增殖和功能活动的影响。结果显示,100μA/cm^2的直流电,可促使成骨细胞内钙离子通道开放,增加细胞内游离钙离子浓度,促进DNA合成,加快成骨细胞的分裂增殖速度。结果证明,钙离子浓度变化是调节成骨细胞功能活动的关键因素,也是电刺激促进骨生长改建的机制之一。The experimental model of in vitro culture and direct current stimulation of isolated rat osteoblast-like cells was established and used for studying the effects of mechanoelectric environments on osteogenesis and bone metabolism. DNA synthesis and Ca^(++) concen- tration in the osteoblast-like cells were measured with an adherent cell analysis and sorting machine(ACAS-570). The resultsshowed that suitable direct current stimulation (e. g. 100μA/cm^2) was effective in opening the Ca^(++) passage in the osteoblast-like cell membrane, increasing the intracellular free calcium ions density, accelerating the speed of DNA synthesis and promoting the proliferation of osteoblast-like cells. It also had a strong effect on the orientation of the stimulated cells. Form the results, it was concluded that introextro-cellular calcium ions metabolism plays a key role in osteogenesis under direct current stimulation.
