摘 要:探讨大鼠肝细胞GSH转运体系在非洲爪蟾卵中表达的可能性。用大鼠肝细胞mRNA注射蛙卵,检测蛙卵转运GSH的活性。结果:注射大鼠肝细胞mRNA的蛙卵,3d后能表达摄取和释放GSH的转运活性,而注射水的蛙卵则不显示这种转运活性。将大鼠肝细胞mRNA进一步分段离心后分别注射蛙卵,发现大鼠肝细胞的两类GSH转运蛋白分别由分子大小为2.0~3.0kb和4.0~5.0kb的mRNA所编码。结论:非洲爪蟾卵可用于检测和克隆大鼠肝细胞GSH的转运蛋白。PURPOSE The possibility of the expression of rat liver GSH transport system in Xenopus laevis oocytes was studied.METHODS Oocytes were injected with rat liver mRNA and GSH transport activities of oocytes were measured.RESULTS The oocytes injected with rat liver mRNA required 3 days to express the activities to release or uptake GSH,while the oocytes injected with water did not show any activities.Using size-fractionation of rat liver mRNA,2 fractions were found to encode the two different GSH transporters,2.0~3.0 kilobase and 4.0~5.0 kilobase mRNAs respectively.CONCLUSIONS Xenopus Laevis oocytes may be used for studying and expression cloning of rat liver GSH transporters.
