绞股蓝总皂甙和人参总皂甙对大鼠急性心肌缺血心脏舒张功能保护作用的比较

摘　　要：目的：评价绞股蓝总皂甙（Ｇｙｐ）对大鼠急性心肌缺血心脏舒张功能保护作用，并同人参总皂甙（Ｇｉｎ）做比较。方法：用ＳＭＵＰ－１６０心功能测定软件，分别测定正常对照，提前给予Ｇｙｐ（ｉｖ，４０ｍｇ／ｋｇ）和Ｇｉｎ（ｉｖ，４０ｍｇ／ｋｇ）组大鼠心脏的舒张功能。结果：Ｇｙｐ（ｉｖ，４０ｍｇ／ｋｇ）对麻醉大鼠基础心功能无影响。Ｇｉｎ（ｉｖ，４０ｍｇ／ｋｇ）可显著降低心室收缩峰压（ＬＶＳＰ）和心率（ＨＲ）（Ｐ＜０．０５或Ｐ＜０．０１），一过性降低心室压最大上升和下降速率（±６Ｐ／ｄｔ＿（ｍａｘ））和心肌纤维零负荷下最大缩短速率（Ｖ＿（ｍａｘ）），对等容舒张期压力下降时间常数Ｔ无影响。结扎大鼠冠脉左前降支即刻引起左室舒张功能损伤并于１０ｍｉｎ达到峰值，表现为等容收缩相Ｔ值显著延长，－ｄＰ／ｄｔ＿（ｍａｘ）和－ｄＰ／ｄｔ＿（ｍａｘ）·ＬＶＳＰ￣（－１）明显降低，心室舒张未压（ＬＶＥＤＰ）则明显升高。冠脉结扎前给予Ｇｙｐ或Ｇｉｎ可显著（Ｐ＜０．０５或Ｐ＜０．０１）拮抗Ｔ值的延长，ＬＶＥＤＰ的抬高和－ｄＰ／ｄｔ＿（ｍａｘ），－ｄＰ／ｄｔ＿（ｍａｘ）·ＬＶＳＰ￣（－１）的降低。结论：Ｇｙｐ和Ｇｉｎ对大鼠实验性心肌缺血心脏舒张功能?OBJECTIVE : To compare the protective effect of gypenosides(Gyp)on left ventricular relaxationwith that of ginsenosides(Gin) during acute myocardial ischemia in rats. METHOD:By using SMUP-160 software,left ventricular relaxation of rat was determined among NS,Gyp(iv,40 mg/kg)and Gin(iv,40 mg/kg)Pretreatedgroups. RESULTS : Gyp(iv, 40 mg/kg ) had no effect on all the cardiac function parameters while Gin(iv, 40mg/kg)caused great (P< 0. 05 or P<0. 01)decrease of LVSP and HR,transient decrease of ± dP/dtmax and Vmax but no effect on T. Left ventricular relaxation was markedly impaired during acute myocardial ischemia in anes- thetized rats with T prolonged,-dP/dtmax and一dP/dtmax·LVSO-1 declined as well as LVEDP elevated signifi- cantly. The impairment of left ventricular relaxation occurred after the onset of coronary artery occlusion andpeaked at 10 min. Gyp and Gin administered prior to occlusion resulted in significant(P< 0. 05 or P< 0. 01) decrease of T, LVEDP and augment of-dP/dtmax and-dP/dtmax·LVSP-1 compared with NS group.CONCLU- SION: Both Gyp and Gin had protective effects on left ventricular relaxation during acute myocardial ischemia inrats while Gin was more effective in ameliorating decrease of LVEDP induced by acute myocardial ischemia thanGyp.
