摘  要:目的探讨补阳还五汤有效部位对大脑局灶性脑缺血后再灌注大鼠脑组织IL—1β和ICAM—1表达的作用。方法采用大鼠大脑中动脉闭塞局灶性脑缺血再灌注模型,检测脑组织IL—1β和ICAM—1的表达。结果脑缺血2 h再灌注46 h后,模型组海马及髓质区IL—1β表达阳性细胞数显著增加(P<0.05,P<0.01),海马及皮质区ICAM—1表达阳性细胞数显著增多(P<0.01,P<0.05);补阳还五汤有效部位生物碱可使髓质区IL—1β阳性细胞数显著减少(P<0.01);苷可使皮质区ICAM—1β阳性细胞数显著减少(P<0.01)。结论脑缺血再灌注后,脑组织IL—1β和ICAM—1表达增加,补阳还五汤有效部位生物碱和苷可分别抑制髓质部位IL—1β蛋白表达及皮质区ICAM—1蛋白表达,提示生物碱和苷可能是补阳还五汤抗脑缺血后炎症反应的部分物质基础。Objective To investigate the effect of active fraction of Buyang Huanwu Decoction (BYH- WD) on cerebral IL-1β and ICAM-1 expression in middle cerebral artery occlusion followed reperfusion in rats. Methods The model rats of cerebral ischemia reperfusion were induced by the method of nylon monofia ment via the middle carotid artery, IL-1β and ICAM-1 expression in cerebral tissue were determined. Results After cerebral ischemia for 2h followed reperfusion in 46h, IL- 1 β positive cells in cerebral hippocampus and medulla increased markedly( P 〈 0.05, P 〈 0.01 ), ICAM- 1 positive cells in hippocampus and cortex increased remarkably( P 〈 0.01, P 〈 0.05), alkaloid extracted from BYHWD could reduce IL- 1β positive cells( P 〈 0.01 ), glgcoside extracted from BYHWD could decrease ICAM- 1 positive cells ( P 〈 0.01 ). Conclusion After cerebral ischemia reperfusion, both IL-1β and ICAM-1 expression are increased, alkaloid from BYHWD could inhibit IL-1β protein expression in cerebral medulla, glycoside from BYHWD could inhibit ICAM-1 protein expression in cerebral cortex. Alkaloid and glycoside may be the partial substance basis of BYHWD antagonizing inflammatory reaction after cerebral ischemia reperfusion.
