摘  要:目的制备重组人干扰素-βSer17(rhIFN-βSer17),并检测其对SARS病毒的抑制作用。方法通过发酵收集菌体,经高压匀浆破碎收集包涵体、有机溶剂抽提、酸沉淀、柱层析纯化重组蛋白,通过细胞病变抑制法检测其对SARS病毒的抑制作用。结果研制的rhIFN-βSer17的纯度超过95%,比活性超过2.0×107IU/mg蛋白,在体外对SARS病毒具有明显的抑制作用。结论已成功制备rhIFN-βSer17,并在体外对SARS病毒具有抑制作用。Objective To prepare recombinant humaninterferon β Ser^17(rhIFN-βSer^17) and study its inhibitory effect on SARS virus. Methods The recombinant E. coli strain expressing interferon β Ser^17 was incubated by fermentation, harvested and broken by high pressure homogenizer,then inclusion bodies were collected,extracted with organic solvent and purified by acid precipitation and gel filtration chromatography.The inhibitory effect of rhIFN-β Ser^17 on SARS virus was studied by CPE assay in vitro. Results The purity of expressed rhIFN-βSer^17 was more than 95% ,and its specific activity was more than 2 × 10^7 IU/mg protein. The rhIFN-β Ser^17 showed significant inhibitory effect on SARS virus in vitro. Conclusion rhIFN-β Ser^17 was successfully prepared and showed significant inhibitory effect on SARS virus in vitro.
