摘  要:超氧基是吞噬细胞呼吸爆发系列酶反应的主要代谢产物,它对抗感染、抗肿瘤、增强机体防御能力有重要意义。用Hypaqule-ficoll梯度离心法从健康人外周血分离的中性白细胞(PMN)与肿瘤坏死因子(TNFα)和多聚不饱和脂肪酸(PUFA)分别或先后温育,用化学发光法(Chemiluminescence,CL)和超氧基歧化酶(SOD)抑制高铁细胞色素C还原法测定被激发细胞的超氧基释放水平,两种测定法获得一致的结果。研究结果表明,包括花生四烯酸(AA)、二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)等PUFA均能明显加强PMN的超氧基释放。经TNFα预处理的细胞,超氧基释放量明显增加。超氧基释放水平与PUFA和TNFα使用浓度呈正相关。Using hypaque-ficoll differential density centrifugation, we prepared polymorpho nuclear nutrophil leucocytes(PMNs)from peripheral blood of healthy adult donors. The cells were pretreated with tumor necrosis factor α(TNFα) of/then treated with polyunsaturated fatty acids (PUFA) . Comparing the two methods of lucigenin-dependent chemiluminescence and superoxide production determined by superoxide dismutase(SOD) inhibiting reduction of cytcthrom C , we have obtained similar results. PUFA including arachidonic acid (AA,20:4),ecosapentaenoic acid ( EPA , 20 : 5 ) , docosahexaenoic acid (DHA , 22: 6 ) induced superoxide release of neutrophils. The cells pretreated with TNF were more sensitive to PUFA for enhanced superoxide generation significantly. Also , we found that the .superoxide levels increased were dose dependent of PUFA and TNFα
