摘 要:人膀胱癌单克隆抗体BDI-1通过异型双功能交联剂SPDP与载阿霉素人血清白蛋白毫微球偶联,制备了免疫毫微球DBI-1-HSA(ADR)-NS。此免疫毫微球在体外对人膀胱场细胞BIU-87及E-J有特异杀伤效应,而对人直肠癌细胞Lovo无杀伤;荷瘤裸鼠体内实验表明,此免疫毫微球可明显抑制人膀胱癌细胞的生长。Immunonanospheres (BDI-1-HSA (ADR ) -NS) against human bladder tumor cell lines were constructed by covalently cross linking the monoclonal antibody BDI-1 with adriamycin loaded human. serum albumin nanospheres via hetero bifunctional crosslinker SPDP.BDI-1-HSA (ADR)-NS exerted the specific cytotoxic effects on the target cells human bladder tumor cell lines BIU-87 and E-J , but not on the non target cells human colon carcinomar cell line Lovo. The results of the inhibitory activity of the immunonanospheres on the human bladder cell lines E-J in nude mice xenograft model ,showed that the immunonanospheres could postpone the time of carcinogenesis and reduce the growth of tumor significantly. The therapeutic effect of the immunononanospheres on the tumor , in nude mice xenograft model , was significantly superior to the normal saline and the mixture of BDI- 1 and ADR-HSA-N S.
