摘　　要：通过实验确定了采用异硫氰酸胍和β-巯基乙醇联合变性,酚、氯仿、异戊醇抽提的方法,从一株次生代谢产物为苝醌衍生物的丝状真菌AL18中提取总RNA。经甲醛变性凝胶电泳分析和紫外分光光度法检测,证实得到了完整、均一的总RNA,为构建此菌的cDNA文库和克隆生物合成关键基因奠定了基础。The method was established for total RNA extracting from a fungus ALl8 producing Perylenequinonoid derivatives photosensitizer using guanidinium isothiocyante and β - mercaptoethanol for denaturing, then using water - satured phenol , chloroform and isoamylalcohole for extracting. The integrity and purification of the total RNA isolated are eligible by using formaldehyde - agarose gel electrophoresis and UV spectrophotometer,so it is available us to construct cDNA library and even to clone the key gene for biosynthesis.
