摘 要:目的:探讨精子优选和精液冷冻对精子顶体完整率的影响。方法:16例精液样本采用Percoll和上游两种方法优选,13例精液标本采用快速和缓慢冷冻处理,采用巴氏染色法分析精子顶体完整率。结果:Percoll和上游两种方法优选后精子顶体完整率均明显高于优选前(P<0.05),两种优选方法间顶体完整率比较差异无显著性(P>0.05)。冷冻后与冷冻前顶体完整率比较差异无显著性(P>0.05),甘油-卵黄-枸橼酸钠(GYC)和甘油两种冷冻保护剂对顶体完整率的影响差异无显著性(P>0.05),速冻和缓慢冷冻对精子顶体完整率的影响差异亦无显著性(P>0.05)。结论:Percoll和上游法优选后精子顶体完整率较优选前明显改善。冷冻过程对顶体完整率无明显影响,速、缓冷冻方法和冷冻保护剂类型对顶体完整率亦无明显影响。Objective To explore the effects of cryopreservative methods and cryopreservative mediums on the acrosome intactness rate of human spermatozoa. Methods Human semen samples from 16 donors were selected by Percoll and swimming up methods, human semen samples from 13 donors were cryopreserved by fast frozen and slow frozen methods. The rate of acrosome intactness were measured and analyzed. Results The rate of acrosome intactness were significantly increased after Percoll gradient centrifugation and swimming up (P〈0.05). The comparison between Percoll gradient centrifugation group and swimming up group showed that the rate of acrosome intactness was not significantly different (P〉0.05). Post-thawing the rate of acrosome intactness was not significantly different(P〉0.05). There were no statistics differences in the rate of acrosome intactness between fast frozen and slow frozen, GYC and glycerol groups (P〉0.05). Conclusion Pet-coil gradient centrifugation and swimming up technique may be widely applied in reproductive medicine. Cryopreservation could not affect the acrosome intactness rate of human spermatozoa. The cryopreservation methods and cryoprotective mediums could not affect the acrosome intactness rate of human spermatozoa.
