摘 要:目的研究中药复方糖肾宁对高糖诱导的肾近端小管上皮细胞转化生长因子β1(TGF-β1)表达和分泌纤维连接蛋白(FN)的影响,探讨其治疗糖尿病肾病(DN)的机制。方法分别采用RT-PCR和ELISA方法检测不同时间下高糖对肾近端小管上皮细胞TGF-β1 mRNA表达和分泌FN的作用,及糖肾宁药物血清对上述指标的影响。结果高糖呈时间依赖性增加肾小管上皮细胞TGF-β1 mRNA表达,高糖培养24h后细胞分泌FN也明显增多,加入糖肾宁药物血清后上述指标均有下降(P〈0.05)。结论糖肾宁抑制高糖诱导的肾小管上皮细胞TGF-β1和FN表达可能是其治疗DN的分子机制之一。Objective Toinvestigatethe effects and mechanism of Tang Shen Ning(TSN)on expression of transforming growth factor-β1 (TGF-β1) and excretion of fibronectin(FN) in renal proximal tubule cells cultured in high-glucose solution. Method Reverse transcription-polymerase chain reaction and enzyme-linked immunosorbent assay was used respectively to detect the expression of TGF-β1 and the secretion of fibronectin(FN)in renal proximal tubule cells cultured in high-glucose solution at differrent time, and the effects of the serm contaimed Tang Shen Ning on the above-parameters were observed. Results The expression of the TGF-β1, mRNA increased with time on high glucose culture. The increased FN levels also could be found after 24h culture in high-glucose solution. After Tang Shen Ning was added were decreased(P〈0. 05). Conclusions TSN could inhibit the expression of TGF-β1 and excretion of fibronectin in renal proximal tubule cells induced by high-glucose solution, and this might be one of the molecular mechanisms to treat diabetic nephropathy.
