摘 要:采用紫外-可见光谱、荧光光谱、微分脉冲伏安法以及紫外-可见光谱电化学法等多种手段,从分子水平研究了紫杉醇对柔红霉素与DNA作用的影响.紫杉醇可以和柔红霉素形成分子间氢键;紫杉醇的长链对柔红霉素糖环以及柔红霉素和DNA所形成加合物的外围有一定程度的缠绕作用,最终使柔红霉素的药效增加毒副作用减小.As a kind of diterpenoida, paclitaxel is a new antitumor drug. The combined use of paclitaxel and anthracyline antibiotic drug such as daunorubicin can gain more remarkable curative effect and less toxicity in the process of clinic treatment. In order to study more about the mechanism of their synergistic action, the influence of paclitaxel on the antitumor effect of daunorubicin under metabolic condition was investigated in this paper by UV-visible spectrophotometry, fluorescence spectrophotometry, differential pulse voltammetry and UV-visible spectroelectrochemistry methods. The results demonstrate that paclitaxel and daunorubicin can form intermolecular hydrogen bonds. Furthermore, paclitaxel's long chain can wind the aglycone of daunorubicin and the peripheral of the adduct formed by daunorubicin and DNA. Consequently, the side-effects of daunorubicin are decreased and its curative effect is enhanced.
