摘 要:通过活细胞计数、3H-TdR掺入法、细胞因子生物活性检测、电镜及免疫组化方法研究了维甲酸对角朊细胞体外增殖分化的影响。结果表明维甲酸能以一种剂量依赖方式抑制人角朊细胞株Cole-16细胞的体外增殖和DNA合成;减少角朊细胞肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)的产生,对角朊细胞超微结构有直接损伤作用;并抑制角朊细胞角蛋白表达。提示维甲酸是角朊细胞体外增殖分化的强有力抑制药物。Using the culture of human keratinocyte cell line Colo-16 cells as a model, the inhibitory effects 0f retinoic acid (RA) on the proliferation and DNA synthesis were observed as determined by viable cell count and 3H-TdR uptake. In the supernatant derived from Colo-16 cell culture treated with RA, the tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)production were significantly reduced.Transmission electron microscopic observation and immunohistochemical technology revealed that the nuclear membrance and organelles of keratinocytes were seriously damaged and the keratin expression as markers of keratinocytes squamous-cell differentiation was suppressed. This study suggests that RA may be a powerful inhibitory drug against the proliferation and differentiation of keratinocyes.
