碘缺乏阻碍大鼠小脑胶质细胞的发育

摘　　要：碘缺乏阻碍中枢神经系统的发育,但对胶质细胞影响的研究却很少。本实验用免组化方法,研究了用严重缺碘粮食喂养大鼠4个月后所生仔代小脑星形胶质细胞的变化,进一步阐明了碘及甲状腺激素影响小脑发育的机制。实验结果表明:实验组母鼠与仔鼠血清 T_4明显下降,甲状腺肿大,小脑分叶程度落后,外颗粒层消失延迟,分子层发育落后;外颗粒细胞层与分子层内 GFAP 免疫性纤维减少。内颗粒细胞层与髓质内 GFAP 阳性胶质细胞与纤维减少,胶质细胞的这种改变直到生后60日龄仍可观察到。这提示:缺碘导致的甲功低下阻碍了小脑星形胶质细胞的发育,进而影响了小脑功能。It is well known that the development of central nerve ststem is retarded by iodine deficiency,but the information on gila development under the condition of iodine deficiency is limited.In this experiment cerebellar astroglia of rat offsprings whose mothers were fed on the food of severe iodine defieieney for 4 months was studied hy immunohistochemistry in order to elucidate the mechanism of effect of iodine and thyroid hormones on eerebellar development further.This experimental results indicated that serum T_4 of offspring and mother rats was markedly decreased in experimental groups,and that there was en- largement of thyroid glands,delayed development of cerebellar lobules,delayed disappearance of external granular layer and retardation of molecular layer of cerebellum in experimental groups.It was shown that positive GFAP staining fibrae in external granular layer and molecular layer of experimental groups was less,positive GFAP astroglia and fibrae in internal granular layer and white matter were decreased. The delayed changes of astroglia were maintained until postnatal 60 days of development of the experi- ment.The results suggested that astroglia development of cerebellum was retarded by hypothyroidism induced by iodine deficiency and cerebellar function was influenced due to the defective astroglia.
