系统性白念珠菌感染宿主预后与肾脏脾脏免疫反应类型的关系

摘  要:目的:探讨系统性白念珠菌病宿主预后与不同类型器官内免疫反应类型的关系。方法:经小鼠尾静脉接种不同剂量白念珠菌以建立系统性白念珠菌病模型,观测小鼠的生存时间,逆转录-多聚酶链式反应(RT-PCR)方法检测小鼠脾脏及肾脏相关细胞因子(IFN-γ、IL-4)的表达,研究小鼠的预后与其脾脏、肾脏内免疫反应类型的关系。结果:无论接种致死量还是非致死量的白念珠菌,脾脏IFN-γ无明显改变,而IL-4表达明显升高,即呈现II型免疫反应;而肾脏在接种非致死量时IFN-γ显著升高,而IL-4无明显改变,即呈现I型免疫反应,接种致死量时IFN-γ无明显改变,而IL-4显著升高,即呈现II型免疫反应。结论:本研究提示系统性白念珠菌病宿主的预后可能主要与肾脏内的免疫反应类型有关,而与脾脏内免疫反应类型无明显相关性。Objective: To determine the relationship between host survival and the type of immune responses in different organs in disseminated Candidiasis. Methods: A murine model of disseminated Candidiasis induced by injection with Candida albicans via the tail vein was established. The survival time was monitored and the expression of cytokines (interferon -γ and interleukin - 4) in the spleen and kidney was detected with RT - PCR. Results: Both nonfatal and fatal doses injected cause a type - Ⅱ immune response in the spleen. In kidney, conversely, nonfatal doses induced a type - Ⅰ immune response and fatal doses induced a type - Ⅱ immune response. Conclusion: In disseminated Candidiasis the host survival is relevant with the type of the immune response in kidney, but not in spleen.
