TLSF_（JM）对PMA诱导Jurkat细胞IL－2Rα链基因表达的抑制作用

摘　　要：本文应用原位杂交和双ＭｃＡｂ夹心ＥＬＩＳＡ方法，探讨了来自ＪＭＴ细胞白血病细胞系产生的抑制因子（ＴＬＳＦ_（ＪＭ））对ＰＭＡ诱导的Ｊｕｒｋａｔ细胞ＩＬ－２Ｒα链基因表达的影响。结果表明：ＴＬＳＦ_（ＪＭ）可明显抑制ＰＭＡ诱导的Ｊｕｒｋａｔ细胞ＩＬ－２Ｒα链ｍＲＮＡ转录，并降低膜表面ＩＬ－２Ｒα链的表达和细胞培养上清中可溶性ＩＬ－２Ｒ（ｓＩＬ－２Ｒ）水平。By using mRNA molecular hybridization and double McAb sandwich ELISA,we observed the effects of TLSFJM on IL-2Rαchain gene expression of Jurkat cells induced by PMA.The results showd that TLSFJM markedly inhibited the expression of IL-2Rα chain mRNA on Jurkat cells and decreased the levels of membrane IL-2R(mIL-2R) and sIL-2R in supernatant.These results suggexted the inhibitory effects of TLSF_(JM) on levels both membrane bound IL-2Rα chain and slL-2R in supernatants are at least partly due to decreased IL-2Rα chain gene expression.
