摘  要:目的观察不同阶段海洛因成瘾大鼠脑内去甲肾上腺素(NE)、多巴胺(DA)、5羟色胺(5HT)及5羟吲哚乙酸(5HIAA)的变化。方法模拟人类吸毒成瘾、治疗;复吸成瘾,再治疗周而复始的模式。按递增法给大鼠染毒,造成动物海洛因成瘾。剖取不同阶段的鼠脑,以荧光分光光度法检测单胺类递质含量。结果各阶段海洛因成瘾鼠脑单胺类递质含量明显增高,5HT代谢物亦明显增高。α银环蛇毒素和美沙酮等治疗均能显著降低单胺递质。结论海洛因成瘾的大鼠,不同阶段的脑内单胺递质明显升高,两种治疗方案均可显著降低单胺类递质、缓解戒断症状。OBJECTIVE To study the changes of NE,DA, 5 HT and 5-HIAA at different periods in the brain of heroin depend ence-rats. METHODS The drug dependence-animals were made by injecting heroin in gradual increasing dosage and the relapse models of heroin addiction were established by a similar way to the human being drug abusers, i. e drug-taking dependence, treatment;relapse dependence, re-treatment again. The rat brain specimen were obtained from different periods and the contents of monoamine transmitters were detected by fluorescence photometric method. RESULTS The contents of monoamine transmitters were increased prominently at different periods in the brain of heroin dependentce-rats, the metabolic products of 5 HT were also increased. Monoamine transmitters were decreased after treatment with both α-bungarotoxin and methadone therapy. CONCLUSION Monoamine transmitters were increased significantly at different periods in the brain of heroin dependence-rats, α-bungarotoxin and methadone therapy could inhibit the increase of monoamine transmitters and relieve withdrawal symptoms.
