摘  要:目的观察吡格列酮(Pio)对血管内皮细胞一氧化氮(NO)和内皮素1(ET-1)分泌的影响.方法不同浓度Pio处理体外培养的血管内皮细胞(ECV304)72 h,硝酸还原酶法和放射免疫法测定细胞上清液内NO和ET-1水平.结果(1×10-8~1×10-5)mmol/LPio可显著促进ECV304细胞NO的释放和抑制其ET-1分泌,作用呈浓度依赖性.结论Pio可促进血管内皮细胞NO释放和抑制其ET-1分泌,这可能是Pio血管保护作用的机制之一.Objective To evaluate the effects of pioglitazone(Pio) on the secretion of nitric oxide and endothelin-1 in vascular endothelial cells. Methods Cultured vascular endothelial cells(ECV304) were incubated with various concentration pioglitazone. The levels of NO and ET-1 in the supernatants of the cultures were assayed by nitrate reductas method and RIA after 72 hours pretreated. Results The levels of NO in the supernatant of Pio treated group(concentration from 1 × 10^-8 to 1 ×10^-5 mmol/L) were higher than these of control group. But the levels of ET-1 in the supernatant of culture cell with same Pio concentration were lower than control group. Conclusion Pio can promote NO release and inhibit ET-1 release, all these maybe one of the mechanisms of Pio effect in vascular protect.
