摘 要:用6个微卫星标记对中国对虾(Fenneropenaeus chinensis)的5个家系进行系谱鉴别和遗传多样性研究.6个微卫星位点中有5个位点是多态的,并在所有家系中都显示了高度的遗传差异.5个家系中,5个多态微卫星位点共发现了30个等位基因,每个位点的等位基因数在5~8之间.实验中共发现了4个家系特异性等位基因:2#家系及4#家系各1个,5#家系2个.根据已知亲本及子代基因型,可推断出5个家系中全部亲本的基因型,据此鉴别各家系.在EN0033位点,可将5#家系与其他4个家系相区别;在RS0859位点,可将3#和4#家系与其他3个家系相区分.因此,EN0033和RS0859标记可用于鉴别5#、3#和4#家系的家系特异性标记.研究表明,用5个微卫星标记,且最少用2对微卫星标记即可鉴别5个中国对虾家系.One of the difficulties in selecting a breeding program is to maintain the parentage information. In order to obtain the survival and growth traits of each family, the progeny from family groups are often isolated and raised in separate tanks until they are large enough for physical tagging. Due to their highly polymorphic, microsateUites have provided an essential tool for identifying parentage relationships among individuals and for obtaining pedigree information in aquaculture selective programs. Six microsatellite markers were used to estimate the kinship and examine the genetic diversity of five Fenneropenaeus chinensis families. Investigating the potential application of microsatellite markers in the parentage determination of Fenneropenaeus chinensis, as well as exploring family unique markers, which could be used in marker-assisting selective breeding and gene localization, was also performed in this study. All the microsatellite loci were polymorphic except locus EN0018. The five polymorphic loci displayed high genetic difference among the five families. The polymorphic information content (PIC) of the five loci was 0.806 5 at locus ENO033, 0.696 8 at RS0683, 0.7287 at RS0859, 0.741 4 at RS062 and 0.741 4 at RS0622, respectively. Thirty alleles were detected in five families tested at five microsatellite loci. The number of the alleles at each locus was between five and eight. There were six alleles at locus RS0683, six at locus RS0859, five at locus RS062 and five at locus RS0622. Four family unique alleles were. found out: one in 2^#family, one in 4^# family and two in 5^# family. Family unique allele was not found in families 1^# and 3^# Sixty kinds of genotypes were acquired. Thirteen genotypes were acquired at locus ENO033, 10 at locus RS0683, 13 at locus RS0859, 11 at locus RS062 and 11 at RS0622, respectively.
